Three healthcare names drew eight analyst actions on Tuesday, the first session after the Labor Day break. Ionis took five of them, Amgen two and BioNTech one. One desk downgraded twice. Six target figures came down and not one was raised.
Ionis Books $268 Million In Quarterly Revenue And Draws Five Analyst Actions
Ionis makes RNA-targeted medicines. Each one is a short strand built to switch off a single gene. Four product lines carry the revenue. Two are its own, TRYNGOLZA and DAWNZERA. The other two are royalties on SPINRAZA and WAINUA, which partners sell. Revenue over the last twelve months was $874 million.
What The Pelacarsen Trial Showed
Novartis released topline Phase 3 results for pelacarsen on 4 September. Ionis discovered the drug and licensed it to Novartis. The Lp(a)HORIZON trial enrolled 8,323 patients with elevated lipoprotein(a) and established cardiovascular disease.
Lipoprotein(a) is a fatty particle in the blood linked to heart disease. Pelacarsen is built to lower it. The primary endpoint combined four kinds of event. They were cardiovascular death, non-fatal heart attack, non-fatal stroke, and urgent coronary revascularisation needing a hospital stay. That endpoint was not met. The drug did lower lipoprotein(a) against placebo. Shreeram Aradhye, the Novartis president of development and chief medical officer, said the findings did not demonstrate that this translated into reduced cardiovascular risk in the overall group. The 4 September release said the data will go to a medical congress.
The FDA approved ZANVASTRO, the Ionis drug zilganersen, on 3 September. It is the first disease-modifying treatment for Alexander disease.

Ionis reported its second quarter on 29 July. Revenue was $268 million, against $452 million a year earlier. The net loss was $115 million, or $0.69 a share. Cash and short-term investments stood at $2.055 billion on 30 June.

David Lebowitz of BMO Capital trimmed his from $61 to $54 at Market Perform. Michael Ulz of Morgan Stanley marked his down from $110 to $104 at Overweight.

Amgen reported its second quarter on 4 August. Total revenue was $10.1 billion, 10 percent above the same quarter a year earlier, and product sales were $9.537 billion. Non-GAAP earnings were $6.29 a share, 4 percent higher. On a GAAP basis earnings were $4.37 a share, against $2.65.
Repatha sales were $953 million, 37 percent higher. Evenity was $714 million, 38 percent higher. Tezspire was $486 million, 42 percent higher. Free cash flow was $3.5 billion in the quarter, against $1.9 billion a year earlier. Amgen guides to full year revenue of $38.2 billion to $39.4 billion and non-GAAP earnings of $22.30 to $23.50 a share.

Two desks acted on Tuesday and neither moved a number. Evan Seigerman of BMO Capital downgraded Amgen to Market Perform from Outperform, holding his $450 target. He wrote that commercial execution is now priced in. Patents running out across the portfolio were part of his case. He also questioned how the obesity candidate MariTide compares with treatments already sold. Carter Gould of Cantor Fitzgerald reiterated Neutral and kept $400.

BioNTech reported its second quarter on 4 August. Revenue was 105.6 million euros, against 260.8 million a year earlier. The net loss was 820.8 million euros, or 3.24 euros a share. Adjusted research and development spending was 477.1 million euros, down from 509.1 million. Cash and security investments were 16.6 billion euros on 30 June.
The company cut its full year revenue guidance to a range of 1.6 billion to 1.9 billion euros, from 2.0 billion to 2.3 billion. It attributed that to softer global covid vaccine demand. On its lead cancer candidate, pumitamig, it said the drug showed encouraging efficacy in first line non-small cell lung cancer. The company added that response rates were high whatever the level of PD-L1, the protein used to sort which lung tumours respond to immunotherapy.

One desk acted on Tuesday. Seigerman downgraded BioNTech to Market Perform from Outperform and took his target from $128 to $105. Comirnaty is eroding more quickly than he had expected, he wrote. He also pointed to the wait for pumitamig data until 2028. A third reason was iNeST, the programme building a cancer vaccine made for each patient, where he now expects less. On pumitamig he wrote that it “looks good, but ‘good’ isn’t differentiated in a crowding field”. He pointed to rival programmes at Pfizer and at AbbVie with RemeGen.
